IL-17 gene variants in Iranian pediatric patients with visceral leishmaniasis |

Background: Visceral leishmaniasis (VL) is a lethal disease mostly caused by Leishmania infantum parasite in the Middle East. The parasite can stimulate the production of IL-17 by Th17. It was shown that IL-17 is strongly and independently associated with the protection against VL. Then, it seems that IL-17 has a complementary role in the human protection against VL. Since the production of cytokines is under the control of their genes, this study aimed to study the probable relationship between VL and the polymorphisms of IL-17 gene. Methods: One hundred and seventeen patients with VL and 146 healthy individuals living in the same area as patients joined this study. All individuals were genotyped for 9 SNPs (rs4711998AG, rs8193036CT, rs3819024AG, rs2275913AG, rs3819025AG, rs8193038AG, rs3804513AT, rs1974226AG and rs3748067AG) which were selected by using the NCBI SNP database and literature using PCR-RFLP method. Results: The distribution of IL-17 rs3819024 and rs3819025 AA genotypes were significantly more frequent in the controls than patients (P=0.0048 and 0.014 respectively) while IL-17 genotypes rs3748067 AA and rs8193038 GG were more frequent in the patients than controls (P=0.000048 and 0.007, respectively). Conclusion: Based on the results the IL-17 rs3819024 and rs3819025 AA genotypes can be considered as resistance factor for VL while rs3748067 AA and rs8193038 GG genotypes may be susceptibility factors for this disease in Iranian pediatric patients.
